Do Delayed Periods Suggest Pregnancy?

My girlfriend went to the gynecologist for a routine check up and she is worried she might be pregnant. They always ask her to pee in a cup but she is unsure if they check for it on a regular visit. She says her body is acting different and she should have had her period sometime in the last week. Would a doctor automatically check her urine for pregnancy?
