It should be ok to take 2 Tylenol PM in advance of a colonoscopy. Tylenol PM contains acetaminophen (pain and fever reducer) and diphenhydramine (the sedating antihistamine that is in Benadryl).
Whenever taking Tylenol in any form, do avoid any alcohol though as the combination is hard on the liver.
Before a procedure, generally NSAIDs such as ibuprofen and naproxen should be avoided, but acetaminophen (Tylenol) is generally ok. And the diphenhydramine should be out of your system by the time you have the procedure. Unless for some reason you are specifically told not to, which should be in the clinic or hospital's patient instructions.
