Skene's gland cyst does not change the testosterone hormone levels in females.
Skene's glands are glands which are homologous with the prostate gland in males.
The role of these glands are in the secretion of fluids during the intercourse.
And these glands have no role in the regulation of hormones like testosterone.
The pain is due to infection of the gland or some other reason like obstruction of the ducts.
Physical examination by the Gynecologist is necessary.
Therefore I advise you to consult the doctor for examination and for treatment.
