Question : My wife, 40, is experiencing too much itching in the anus hole. Also has some itching in the vagina area. Please suggest.

There is a possibility of worm infestation. Give her tablet albendazole 400 mg single tablet in night and tablet cetrizine daily in night for 5 days
This should help her.
If there is no response then it might be a hormonal imbalance and she would require gynaecologist consultation.
