Question : The skin around the testicle area is noticeably red and itchy. I sprayed Benadryl on the area and it significantly burns for several minutes before subsiding.

I read your question and i understand your concern. Itch and redness on scrotal area responding to benadryl is likely a jock itch or some other condition like dermatitis, eczema etc . It needs hygiene measures, keeping the scrota free of hair, Apply some topical hydrocortisone over it, use some antifungal creams as well. Maintain good coital habits, refrain from intimacy for a while. Wear loose undergarments to make sure groin area do not sweat , cotton undergarments will make sure of that.
