Black spot on the penis can be due to penile melanosis, melanocytic naevus, drug reaction or following injury to penis.

Seems to be melanotic macule

Detailed Answer:
On looking at the picture it seems to be melanotic macule which is a benign condition and needs no treatment.

If treatment is required for cosmetic reasons you can go for laser treatment.
