Brief Answer:
I can empathize with you

Detailed Answer:
The recommendations are that we get 150 minutes at least of exercise per week, but a good combination of stretching, cardio and strength training is great for building muscle and losing weight.
Has your doctor advised you that you have arthritis? Your knee joint pain reminds me of arthritis and in that case a simple knee brace might assist in stabilizing the knee joint and consuming omega-3 fatty acids frequently helps with the arthritis.

Sitting in one place and tightening up sounds very much like you are not stretching adquately and maintaining your flexibility. Please stretch before and after all exercises. A good wt. loss kcal amount is 1200 kcal per day with a lowering of high glycemic carbohydrates, increasing fiber, eating lean protein, eliminatinhg most saturated fat and trans fat emphasizing the use of olive oil, nuts, and such.
