Ultram is the trade name for tramadol which is a weak opiate medication.

Glucosamine is a joint nutritional- it helps to repair cartilage in the joints.

It is not necessary for them to be combined but it is possible for the glucosamine to augment (help along) the tramadol.

Tramadol does not interfere with bone cartilage , this was concluded in a medical review document (can provide link if needed).
Since glucosamine helps with cartilage, it is possible that the combination would be helpful
