What Causes A Missed Period With Negative Pregnancy Test?

my wife is not getting her period for 3 months now, and she is getting negative on pregnancy test we are using clear blue for the test. she never missed a period in her life before she is 26. she is worried and I am too. any help you can do? and we are trying to have baby for last 6 months
