Hi, I have gone through your question and understand your concerns. Depo provera injection has side effects of irregular spotting, prolonged amenorrhea or absence of periods till one is on this injection. If your urine pregnancy test is negative, then chance of pregnancy is not there, the irregular spotting could be due to adverse effects of depo provera, or can be due to local lesions in cervix or vagina including cervical cancer. I would suggest you to consult an OBGYN specialist to get properly examined, investigated and treated accordingly. Hope you found the answer helpful. Wishing you good health. Dr Deepti Verma
