It should be used after mifepristone for intrauterine pregnancy (USG should be done prior to use) under the supervision of gynecologist. Following are the causes of absence of period : inadequate dose and improper route of application, ectopic pregnancy. Now I advice you to consult the Gynecologist and go for USG scan to know the pregnancy status. You have to take extra dose of misoprostol under the observation if intrauterine pregnancy will be there. If misoprostol fails then intrauterine pregnancy should be terminated by surgical process like suction evacuation or D & C.
