Question: My wife just noticed a 1" X 2" hard lump on her upper front thigh, nearer the inside. No pain however.

She has history of cancer. She is not diabetic nor does she have any ulcers or other symptoms.

I'm sorry, I mistyped. She has no history of cancer. About 6 to 8 weeks ago, she had a terrible viral head cold with no fever. The worst I ever saw & even got into her eyes. Not given any antibiotics by her doctor since she had no fever.

Isn't it true that there is a high % of false positives from FNAC's. (I assume that is a needle biopsy.) What is the best and accurate biopsy?

Yes, definitely biopsy is better but its invasive and little morbid compared to fnac.

If there's something abnormal on fnac then next step is biopsy.
